Abstract

A ZIC-HILIC glycocapture resin with a dedicated buffer
system was successfully used for highly reproducible
enrichment of glycopeptides in a tryptic digest. The
resulting glycopeptide profiles were rapidly and
reproducibly analyzed by ESI ion trap MS using an Advion
nanomate. LC-MALDI-MS/MS of the glycopeptides
allowed identification of peptide sequences as well as
glycan structures from single MS/MS spectra and thus an
in-depth analysis of all glycosylation sites of a protein in a
single run.
