Abstract

 

There is an urgent need for new and better biomarkers
for use in cancer diagnosis and therapy decision-making
and for delivering new insights into biological processes.
Conventional proteomics approaches have addressed
this need for quite some time. MALDI molecular
tissue imaging has recently emerged as a fully mature
proteomics tool for the direct determination of protein
expression in tissue. This study presents an integrated
workflow – based on Bruker’s PRIME (Proteomics through
Integrated MALDI and ESI) solution – for biomarker
discovery and identification in a complete Top-down
proteomic strategy. The strategy begins with tissue
screening by MALDI imaging, and is using statistical tools
to flag discriminatory proteins. Proteins are identified by
ETD/PTR analysis in the amaZon ion trap. The breast
cancer study presented here reveals a novel putative
biomarker which may become relevant for therapeutic
decision-making.
