In recent years MALDI imaging has become a widely accepted technology in proteomics research and starts to draw increasing interest in clinical research. MALDI imaging is the only molecular imaging technique that does not require preselection of a molecular target, but compared to other molecular imaging modalities its resolution is quite low. The improvement of the spatial resolution is therefore of major importance in the field of MALDI imaging. Here we discuss the factors that are critical for high resolution MALDI imaging, such as matrix application and laser technology. We present MALDI images of proteins that show biologically meaningful structures at 20µm resolution, which is close to cellular resolution and represents the current state-of-the-art. |
