Labeling prior to fractionation

Successful proteomics projects require protein fractionation resulting in a higher sensitivity and proteome coverage.
The new ICPL™ technology (ICPL = Isotope Coded Protein Labeling) supports this promising strategy since it allows protein labeling before fraction and thus maintains the quantitative ratios.

Workflow

  • Cell lysis and ICPL labeling of the different cell states.
  • Combination of both samples.
  • Optional reduction of complexity on protein level, e.g. by 2D GE, PAGE, LC or CLINPROT magnetic beads.
  • Hyphenation of fractionation strategies is possible.
  • Enzymatic digestion of proteins.
  • Optional reduction of complexity on peptide level by LC.
  • Mass spectrometry (Quantitation, Identification) and automated statistical analysis.

The powerful ICPL protein quantitation label technology is now available with four labels:  Serva ICPLTM Quadruplex PLUS (ICPL-4plex Kit)!

The systems allows the simultaneous quantitation of four independent proteome samples by stable isotope protein labeling. The ICPL- technology provides an unmatched dynamic range for protein identification and quantification due to its potential combination with intact protein fractionation steps.

Quantitative MS analysis of proteomes

In contrast to other labelling techniques the ICPL-4plex Kit permits Top-Down proteome analysis using 2D gel or the  GeLC workflow that combines SDS-PAGE protein molecular seizing with LC-MS basis using ESI-MS or MALDI-MS. All approaches that work on a truly proteomics scale use protein pre-fractionation utilizing LC, PAGE, 2D-GE or magnetic beads. Protein fractionation is compatible with the ICPL-technology, as quantitative ratios remain constant throughout any fractionation step. This reduces complexity in each protein fraction, thus increasing the dynamic range of protein detection and quantification and eventually the number of identified proteins. A high sequence coverage of labelled peptides provides a sound statistical base for automatic protein quantification avoiding excessive manual validation efforts.

ProteinScape Integration

ICPL  technology is fully integrated into ProteinScape, Brukers comprehensive bioinformatics platform. ProteinScape stores and manages all relevant data for all types of proteomics projects and delivers reports for publications following the HUPO/PSI publication guidelines.
