Abstract |

The offline coupling of liquid chromatography (LC) to MALDI mass spectrometry (LC-MALDI) is an increasingly used approach to analyse complex protein samples. Major advantages are the reduction of complexity and selection of precursor ions for the MS/MS data acquisition without any time constrains. This allows the intelligent and non-redundant MS/MS analysis based on the preceding MS data, resulting in a high protein identification success. The LC-MALDI analysis of a complex lysate of Corynebacterium glutamicum ATCC13032 allowed the definite Mascot identification of 454 non-redundant proteins, approximately 50% of the bacterium’s proteome. |
